Enzymatic
Assays Reaction Rate Monitoring |
Enzymatic activity or substrate concentration are assayed in FIAlab-on-valve system by reaction rate measurement monitored in real time. Optimization of reaction conditions to Michealis-Menten kinetics can be carried out automatically by varying substrate / enzyme concentrations until the pseudo first order reaction rate conditions are identified. This is achieved by selecting the stopped flow interval at which the substrate/enzyme concentration gradients overlap to the desired degree.
For routine assays such as of glucose, lactate, glycerol, and ethanol, use of prepackaged reagent kits ( such as marketed by Sigma ) is recommended. It is generally straightforward to modify these assays for the use in FIAlab-on-valve system. A substantial saving of reagents is achieved. Since clinical assays have already been scaled down within the existing instrumentation, the reduction of reagent consumption is not as dramatic as achieved for the manual methods, yet a typical reagent kit will yield at least ten times more assays than suggested in manufacturers protocol for a conventional clinical analyzer.
UV-VIS Spectrophotometry as well as Fluorescence detection, integrated into FIAlab-on-valve instrument allows a wide variety of enzymatic assays be carried out by the same apparatus.. Since reaction rate measurement is controlled and evaluated by FIAlab software the calibration and assays can be fully automated.
IMMOBILIZED ENZYMES can be used in LAB-ON-VALVE system in a unique configuration, so called Bead Injection mode. For study of enzyme immobilization, or when using enzymes not stable in solution, Bead Injection technique provides an unique opportunity as a research tool.
TEMPERATURE CONTROL,
essential for enzymatic assays, is presently not built into the FIAlab
system. In favorable laboratory conditions the ambient temperature
is often sufficiently constant, to allow assays to be carried out successfully
bu using periodic calibration with known standards. However,
the ab-on-valve instrument has small overall dimensions that allow
the entire system to be placed into a moderate size incubator.
